I have Sail up and running, all gold.
Now I want to add another card with a couple of FXS, will it loose my actual configuration?

Depends what you mean.  You will have to do an initialise/probe to add the extra cards.  If you don't have any cards then you will lose nothing.  If you do have cards then they will be automatically redefined when you do your new probe.  You will lose their existing routings if they are analogue cards so you will  have to re-enter them.  Other than that, no change.

Stupid question:
what's the easiest way for me to re-do the settings? Write down and re-do manually? Or is there something easier?

Depends which settings you mean.  There really aren't any settings to change for Digital circuits so it only applies to Analogue.  After the initialize and probe your trunks/lines will be recreated in trunks and extensions.  AS far as your trunks are concerned you will have to route them again, which shouldn't take more than a minute or two.  For extensions you may need to change the extension numbers which Sail has allocated to them if they are different this time.  That's it to be honest.  I personally would just take a screen print of them before I did the init/probe.
