Hi, what port is used when accessing the webmail in SME server ? can anyone tell me how I can specify which port a browser should connect to reach the webmail on my sme server.
so they would have to type http://mydomain/webmail:port/

Well it's done over https, so it could be port 443 (standard) but putting your browser to https://mydomain.com/webmail should be enough

You do not need to specify a port to access the sme webmail server in a web interface environment. All you have to do is http://yourserver/webmail and that should be fine.
