I am interested in installing a few GLPI plugins. Looking on the GLPI site, especially the one at http://glpi-project.org/wiki/doku.php?id=en:plugins there should be a "plugins" option in setup - there isn't.
On my server, I see the directory exists in /opt/inventory/glpi/, but there's nothing in it.
Do I need to do anything to enable plugin functionality?

Just extract and upload (with winSCP for example) the plugins you want in the directory /opt/inventory/glpi/plugins, and the "plugins" menu will appear in GLPI interface.

That's what I was hoping, Daniel, but it seemed so ... optimistic. ;-)
Are there any plugins you're using that are worth checking out? I'm specifically interested in a way to manage/change passwords by groups rather than have to go to each machine individually.

Well, That really depend on what you need.

I don't understand which password you want to change

I am hoping that GLPI has a plugin so that I can change the administrative password on 60 machines at once, rather than me having to go to each machine.

If you have a domain setup (which I would do if I was maintaining such a number of computers), you do not need a local administrator account, you should be able to gain administrator access by login in as admin user on the PC when attached to your domain.
