I am really a newbie to this.... i have a server running sme (not sure which version) and noticed that there is  only have 3Gb left on the HDD's (mirrored). I noticed a few Users have email accounts that show between 10-20Gb how do i clear this??

I am sure its simple but i have tried to delete users and still the space is taken on the server but backup shows less on the amount.

Any help would be appreciated

a simple fix would be to add a quota against them. In server manager > Quotas. This should stop them from getting any more mail until they reduce it under the limit. If they go over the limit they'll receive an email telling them to get it sorted. That's the easy way, or did you want to go in an manually look for stuff to delete?

i have tried to apply a quota but it does not free up server space. it still has the "Full" amount shown how would i delete it manually ... mind you i have one sitting at 7gb and i know their email box is nearly empty????

If you add a quota you set a quota for uploading files from the time you set the quota. The files that are already there and are over the quota limit aren't deleted. You'll have to delete the files to reach under the quota and the problem is solved. Do this by ssh into your server and access  users home folders. Or tell them that they won't get mail untill they delete the files.
